Axially overlapped multi-focus light sheet with enlarged field of view

چکیده
Light sheet fluorescence microscopy provides optical sectioning and is widely used in volumetric imaging of large specimens. However, the axial resolution lateral Field View (FoV) system, defined by light sheet, typically limit each other due to spatial band product excitation objective. Here, we develop a simple multi-focus scheme extend FoV, where Gaussian can be focused at three or more consecutive positions. Axially overlapped multiple sheets significantly enlarge FoV with improved uniformity negligible loss resolution. By measuring point spread function fluorescent beads, demonstrated that obtained has 450 μm maximum FWHM 7.5 μm. Compared conventional single-focus one, displays ability over full when cells zebrafish.

Reconstructing Dense Light Field from Multi-Focus Images Array for Virtual View Synthesis
This paper presents a novel method for synthesizing a novel view from two sets of differently focused images taken by an aperture camera array for a scene of two approximately constant depths. The proposed method consists of two steps. The first step is a view interpolation to reconstruct an all in-focus dense light field of the scene. The second step is to synthesize a novel view by light fiel...
متن کاملLight-sheet microscopy with digital Fourier analysis measures transport properties over large field-of-view.
Using light-sheet microscopy combined with digital Fourier methods we probe the dynamics of colloidal samples and DNA molecules. This combination, referred to as selective-plane illumination differential dynamic microscopy (SPIDDM), has the benefit of optical sectioning to study, with minimal photobleaching, thick samples allowing us to measure the diffusivity of colloidal particles at high vol...
متن کاملMulti-view Geometry Estimation for Light Field Compression
Geometry-aided light field compression requires accurate geometry for the efficient representation of the light field image data. In this work, we propose a method to directly estimate a geometry model from the light field images, such that it maximizes the compression efficiency. Previous work on this problem uses least-squares to solve a set of optical-flow based equations. This approach suff...
متن کاملPupil filters for extending the field-of-view in light-sheet microscopy.
Pupil filters, represented by binary phase modulation, have been applied to extend the field of view of a light-sheet fluorescence microscope. Optimization has been used, first numerically to calculate the optimum filter structure and then experimentally, to scale and align the numerically synthesized filter in the microscope. A significant practical extension of the field of view has been obse...
متن کامل3d Two-photon Digital Scanned Laser Light-sheet Microscopy with Large Field of View Using One-side Illumination Beams
Two-Photon Selective Plane Illumination Microscopy (2p-SPIM) was recently demonstrated to be an interesting alternative to its commonly used single-photon counterpart [1]. Although this technique is ideal for accessing larger penetration depth in biological samples it is not optimal in terms of efficiency of fluorescence excitation due to the reduced intensities involved [2]. To overcome this l...
